I just got a new blade and it has an anatomic handle. Are there any disadvantages on using an anatomic handle ?

The shape of the handle doesn't effect performance of the blade. It is just a matter of comfort in your hand.
There are 4 main types of handles, the Anatomic which has a bump in the middle of the handle to try to fit into your hand shape, the Straight handle, the Flared handle and the Conical handle.
Make sure you buy a table tennis bat that is suitable for your level and allows you to develop your technique. For your first bat we recommend the PingSkills Rook, and when you upgrade to a custom bat we recommend the PingSkills Touch with Mark V or Rakza 7.

Would the different handles effect the overall weight of the blade?
I don't think they do but others may be able to shed some light on this.

There will be slight differences in weight between a straight and a flared or anatomic handle for the same blade reference, but you should not really feel it, since this weight delta is in your hand, on the "good side" of the blade's center of gravity. You will feel it much more if you put heavier rubbers on the same blade for example, because that added weight will mostly be on the "bad side" of the center of gravity. Some players even add weights at the extremity of the handle to improve their racket balance.
There are also so types of blades that are available in "filled handles" and "hollow handles" ("Senso" system for Donic). The hollow handles will be lighter, but the center of gravity will be further away from your hand. Some like "nose heavy" blades, and others don't.
